摘要:近年来,量子点(QDs)作为一种新型的纳米荧光材料,引起了国内外研究者极大的关注。量子点用于探索生物领域,生物体成像,分子检测等各方面起到越来越重要的作用。为了进一步提高量子点的荧光稳定性、降低其毒性,本论文以乙酸锌,N-乙酰半胱氨酸(NAC),氨水,正硅酸乙酯(TEOS)为原料,在pH=10.5的条件下对CdSe/ZnS QDs进行包覆,成功合成出CdSe/ZnS@ZnS-SiO2 QDs。通过紫外光谱、荧光光谱对所合成的量子点进行表征。采用荧光光谱法测定了CdSe/ZnS@ZnS-SiO2 QDs体系中加入H2O2的荧光强度,结果表明不同浓度的H2O2有效猝灭了体系的荧光,当向体系中加入还原型谷胱甘肽和谷胱甘肽过氧化物酶后,CdSe/ZnS@ZnS-SiO2 QDs-H2O2体系的荧光逐渐恢复。这将大大提高了量子点在生物分析中的应用价值。因此,本论文进一步为量子点在生物体内的检测与分析提供了新的研究思路。58073

 Application and Research of Quantum Dots in Biological Detection

Abstract: In recent years, as a new type of nano fluorescent material, quantum dots (QDs) have been found. It has aroused great concern of researchers at home and abroad. QDs are becoming more and more important in the field of biology, biological imaging, molecular detection and so on. In order to further improve the stability of QDs and reduce their toxicity, in this paper, CdSe/ZnS QDs will modify whit zinc acetate, NAC, ammonia, TEOS in the conditions of pH=10.5, and synthesized CdSe/ZnS@ZnS-SiO2 QDs. The QDs were characterized by UV and fluorescence spectra, and by fluorescence spectrometric method for the determination of the fluorescence intensity which H2O2 is added to the CdSe/ZnS@ZnS-SiO2 QDs system, the results showed that different concentrations of H2O2 quenching the system of fluorescence, when system to join also reduced glutathione and glutathione peroxidase. CdSe/ZnS@ZnS-SiO2 QDs-H2O2 system of fluorescence recovered gradually. This will increase the value of QDs in biological analysis greatly. Therefore, this paper provides a reference for further study of the application of QDs in biological analysis and detection.
